Peter Parker/Spidey
Though Spider-Man (Tobey Maguire) is finally recognized as a hero (all it took was saving the world ... twice), he has a new worst enemy: himself. When an alien symbiote stains his suit black, it brings out uncharacteristic rage and aggression.
Mary Jane Watson
The love of Spidey's life may now know his favorite hobby, but that hardly erases tension between Mary Jane (Kirsten Dunst) and Peter. She's a perennial target of his adversaries, though it doesn't seem to bother her too much. But her man's superego? That can cause a lover's quarrel.
Venom/Eddie Brock
There's a new hotshot photographer at the Daily Bugle, and he's spiky-haired slickster Eddie Brock (Topher Grace). A profession isn't all he shares with Peter: He also gains arachnid powers ... and picks up an alter ego in the process, Venom. Of course unlike Spidey, Venom is pure evil.
Flint Marko/Sandman
Remember Uncle Ben's heartbreaking demise in the first 'Spider-Man'? You're looking at the killer, Flint Marko (Thomas Haden Church), who becomes the shape-shifting Sandman. Though Sandman seeks forgiveness, he's still No. 1 on Spidey's Most Wanted list.
Harry Osborn/
New Goblin
Peter's old buddy Harry (James Franco) is still seething over the death of his father, and it's no secret he blames Spider-Man. That bitterness drives his full-blown transformation into villainy as New Goblin, who fights Spidey in the air on a kick-ass glider called the Sky Stick.
Gwen Stacy
The daughter of Police Captain Stacy (James Cromwell), Gwen Stacy (Bryce Dallas Howard) is Peter's lab partner at Empire State University. Some might say she's smitten with Parker; but Eddie Brock obsesses over her. Gwen suffered a tragic death in the comic: Will she survive Sam Raimi's version?
Aunt May
Is there anyone sweeter in the comic-book-movie universe than sweet old Aunt May? Negative. May Parker (Rosemary Harris) returns for the third adventure, and shares some tender moments with Peter, including one scene where he admits plans to propose to Mary Jane.
